Identification of Lef1-dependent genes in zebrafish and mouse hypothalamus

We performed RNA-seq with dissected control and Lef1 mutant hypothalami from 3 days post-fertilization (3dpf) zebrafish, embryonic day 14.5 (E14.5) and postnatal day 22 (P22) mouse. We identified Lef1-dependent genes involved in an anxiolytic function in both species, but those genes are different between zebrafish and mouse. Hypothalamic transcriptome of control and Lef1 mutant zebrafish and mouse at different stages were generated by deep sequencing, in triplicate.

我们对受精后3天(3 days post-fertilization, 3dpf)斑马鱼、胚胎第14.5天(embryonic day 14.5, E14.5)及出生后第22天(postnatal day 22, P22)小鼠的解剖对照与Lef1突变下丘脑样本进行了RNA测序(RNA-seq)。我们在两个物种中均鉴定出了参与抗焦虑功能的Lef1依赖型基因,但斑马鱼与小鼠的此类基因存在差异。本研究通过深度测序并设置三次生物学重复,获取了不同发育阶段的对照及Lef1突变斑马鱼、小鼠的下丘脑转录组数据。
