The somatosensory system processes multimodal somatosensory information and acts as a critical hub for organisms to detect potential dangers and sustain survival. We integrated techniques including sparse labeling, fMOST imaging, and single-neuron tracing to map the single-neuron projection profiles of 785 cervical spinal cord projection neurons and 1463 intracerebral relay neurons receiving spinal inputs in mice. These relay neurons are distributed across the thalamus, parabrachial nucleus, motor region of the superior colliculus, hypothalamus, periaqueductal gray, and medulla oblongata. We identified 19 subtypes of spinal cord projection neurons and 34 subtypes of relay neurons. We constructed a connectivity network map of the ascending somatosensory information pathway. Through in-depth analysis of this network, we systematically deciphered the anatomical architecture of the spinal ascending pathway, revealed the organizational principles of the somatosensory system, and laid a solid foundation for subsequent studies on the neural circuit mechanisms of somatosensory information processing.
