The PROFAST pilot study was a double-blinded randomized placebo-controlled trial conducted in Auckland, New Zealand. The primary aim was to determine whether daily probiotic supplementation during a 12 week 5:2 intermittent fasting program could improve glycaemia and reduce weight among adults with prediabetes. 33 participants were randomized 1:1 to receive either probiotic capsules containing Lactobacillus rhamnosus HN001 at a dose of 6x10^9 CFU/day or placebo capsules. Clinical assessments (including stool sample collection) was performed at baseline and at end of the 12 week program. Stool samples underwent shotgun metagenomic sequencing to assess microbiome composition and metabolic capacity.
