To investigate the role of the early growth response gene product (Egr1) in APAP-induced liver injury, liver specific Egr1 deletion mice (Egr1LKO) and their control littermates (Egr1fl/fl) were challenged with 300 mg/kg APAP. Adenovirus encoding Egr1 (Ad-Egr1) was injected into Egr1fl/fl and Egr1LKO mice via tail vein to overexpress Egr1 prior to 300 mg/kg APAP administration. After 12 h post APAP challenge, livers were taken from above mice. Analysis of Egr1-induced changes in global transcriptomic profiles by RNA-seq were performed to investigate the role and mechanism of Egr1 in APAP-induced liver injury.
