Physical Phase Transition of Mineralocorticoid Receptors Determines Preservation Quality of Donor Hearts for Transplantation

We examine the single nuclear RNA profile in the human donor heart at baseline and 10 hours of storage in HTK with/without canrenone. We also analyze the genomic occupancy of MR in these conditions. For snRNA-seq, single nuclei was isolated from human heart tissues and subject to sequencing using 10x platform. For Cut&run seq, single nuclei was isolated from human donor heart and fixed with 0.1% formaldehyde for 1 minutes. The cut&run assay were then performed using Epicypher Cun&run kit.
