Chemotherapy-induced bone loss depends on cellular senescence and the cell secretory phenotype. To investigate the factors secreted in the senescent microenvironment we have defined a mice model o premature senescence and studied the secreted genes that were differentially expressed in the senescent microenvironment. Overall design: To define the chemotherapy-induced senescence model, 15-week-old wildtype C57BL/6 male mice were randomly assigned to Control or DoxoR groups. At day 0, mice were injected intraperitoneally with vehicle (saline solution) or 10 mg/kg doxorubicin. 30 days after the treatment mRNA of tibiae and femur (without bone marrow) of 4 Control and 4 Doxorubicin was isolated.
