Effects of Immunecomplexes on primary human monocytes and their differentitation towards osteoclasts

primary human monocytes were isolated from peripheral blood using Stemcell CD14 magnetic cell seperation and incubated over night with 10ng/ml M-CSF and then differentiated with 100ng/ml RANKL for 24hrs in the absence or prescence of 25 mg/ml SIC (immunecomplexes consisting of SpA and IgG) or the appropriate concentration of IgG or SpA alone. this study includes biological triplicates for each condition (3 independent buffy coats)
