QuantSeq/3' targeted seq supporting study Probing 3’UTRs as modular regulators of gene expression

The goal of the study is to analyze the function of 3'-UTR motifs in the non-native context. Several 3'-UTR motifs predicted to have stabilizing or destabilizing effects on mRNA were inserted into terminators controlling the expression of reporter mCherry gene in following contexts: pRps3-mCherry-tRps3, pPgk1-mCherry-tRps3, pTsa1-mCherry-tTsa. 3'-end sequencing was used to measure the levels of expression of reporter gene and distribution of poly(A) sites.本研究旨在分析非原生背景下3'非翻译区基序（3'-UTR motif）的功能。研究人员将若干经预测可对信使RNA（mRNA）发挥稳定或去稳定作用的3'非翻译区基序，插入至调控报告基因mCherry表达的终止子中，所用表达背景依次为pRps3-mCherry-tRps3、pPgk1-mCherry-tRps3与pTsa1-mCherry-tTsa。随后采用3'端测序（3'-end sequencing）技术，对报告基因的表达水平及多腺苷酸化位点（poly(A) site）的分布特征进行检测。
