The effect of DNMT3A associated clonal hematopoiesis in periodontitis and arthritis

Lethally irradiated CD45.1 mice were transplanted with either 10% Dnmt3aR878H/+ CD45.2 BM cells and 90% WT CD45.1 BM cells (10% Dnmt3aR878H/+ BMT group) or with 10% Dnmt3a+/+ CD45.2 BM cells and 90% WT CD45.1 cells (10% Dnmt3a+/+ BMT group). After 12 weeks, both groups of recipient mice were subjected to LIP for 5 days or CAIA for 7 days, and dissected gingival and synovial tissue were processed for FACS sorting of CD11b+ and CD3+ cells, which were further sorted into CD45.1+ and CD45.2+ cells, followed by scRNA-seq.
