The main goal of this project is to investigate the formation of the non canonical secondary DNA structures G-quadruplexes (G4s) in the mitochondrial DNA (mtDNA) in human cell lines and to disclose their role in the development of mtDNA maintenance disorders.In this study, we developed a new tool to specifically visualize and map G4s in the mtDNA of living cells. Using an engineered mitochondrially-targeted G4-binding protein, we established a mtG4-ChIPseq method that enables the determination of G4 formation in the mitochondrial genome of human cells.
