<p>This dataset contains the experimental data and analysis scripts supporting the findings presented in the manuscript: <strong>"Molecular mechanics of smooth muscle contraction and relaxation modulated by caldesmon."</strong></p>

<h3>Dataset Components</h3>
<ul>
<li><strong>Analysis Code:</strong> Provided as a single <code>.zip</code> file containing Python scripts for data extraction, mathematical modeling, and statistical analysis. Includes a detailed ReadMe file.</li>
<li><strong>Experimental Data:</strong> Uploaded as separate <code>.zip</code> archives (organized by experiment). These archives contain raw microscopy videos and intermediate .mat files generated by the MATLAB-based IVMA analysis software.</li>

<h3>Directory Structure & Experimental Descriptions</h3>
<p>The data and scripts are organized into the following functional directories based on the experimental assays performed:</p>
<ul>
<li><strong>actin_biotinylation_and_CaD:</strong> In vitro motility assays (IVMA) investigating the effects of actin biotinylation on velocity and motile fraction in the presence of CaD (<strong>Figure S2</strong>).</li>
<li><strong>analytical_estimation_of_dePhos_rate:</strong> Mathematical estimation of myosin dephosphorylation rates with and without caldemson (CaD) (<strong>Figure S3</strong>).</li>
<li><strong>mixture_motility_assays (Parts 1 & 2):</strong> Assays using varying ratios of phosphorylated and unphosphorylated myosin with and without CaD. This includes rectangular hyperbola fitting and AIC model comparisons (<strong>Figure 5, Table 3, and Table S2</strong>). <em>Note: This data is split into two parts due to file size limits.</em></li>
<li><strong>MLCP_injections_IVMA:</strong> IVMA experiments during MLCP-induced myosin dephosphorylation (<strong>Figure 4 and Table 2</strong>).</li>
<li><strong>MLCP_injections_LTA:</strong> Laser-trapping (LTA) experiments measuring force-decay and force-rise kinetics, including T_Hold calculations during MLCP-induced relaxation with and without CaD (<strong>Figures 1-3, Table 1, and Figure S1</strong>).</li>

<h3>Technical Notes</h3>
<p>The <code>.mat</code> files included in the data packages were generated using the MATLAB-based <em>In Vitro Motility Analysis (IVMA)</em> software previously described by <em>G. Ijpma et al. (J Muscle Res Cell Motil 39, 153–162, 2018)</em>. The Python scripts provided here process those outputs for final statistical analysis and figure generation.</p>
