We used a multi-omics approach to study the impact of miR-223 in the RAW cell line. We evidenced changes linked to cell death, histone acetylation, bone remodeling, RNA regulation. Changes in miR-223 expression altered the metabolic profile of cells including nucleotides. miR-223 impacted NF-kB levels, macrophage differentiation and osteoclastogenesis. Cell culture was performed in the same conditions for all techniques used. First, RAW 264.7 cells were seeded in 6-well culture plates (Corning) or Petri Dish (10 mm2) at 10,000 cells/cm2, grown until confluency, and transfected for 48 h with irrelevant scramble (Sbl), anti-miR-223 or pre-miR-223 (Qiagen).
