validation of preparation for ampricon sequencing by MiSeq about soil samples. soil metagenome

Project's ultimate goal is structual analysis for bacteria,fungus,protist and nematode about soil. We must extract DNA from soil samples to a maximum extent for structual analysis. Usually we extract DNA from 500mg soil samples by standard method.It is important that how much the result of one DNA extraction show the whole sample's structure. With this aim,we did requirement study for frost shattering before DNA extraction.
