Ingenol mebutate (IM), a therapeutic agent for actinic keratosis, has been reported to possess antitumor activity for pancreatic cancer. However, its mechanism of action is unclear. In this study, we investigated the mechanism of cell growth inhibition against Panc-1, a drug-insensitive pancreatic adenocarcinoma cell line of IM, by RNA-sequencing. The half maximal (50%) inhibitory concentration of cell viability in Panc-1 with 72 hours of IM exposure was 43.1 ± 16.8 nM, but the viability increased in a concentration-dependent manner above 300, and the cell viability sharply decreased again above 30 µM. Thus, this directory includes mRNA sequencing data in Panc-1 cell lines exposed to IM 7.5nM which begins to inhibit cell proliferation, or IM 10uM which begins to cancel the proliferative effect, or DMSO.
