This study was performed to clarify the difference in the bacterial composition between denture and dental plaques. Denture plaque is a multispecies biofilm that grows on the resin surface of removable prosthodontic devices (dentures), and is closely associated with denture-related stomatitis. Methods: Thirty denture plaque and 16 dental plaque samples were collected from 18 denture wearers (mean age, 80.3 years) with normal systemic conditions. After DNA extraction, a meta 16S rDNA amplicon library was constructed using universal PCR primers 341F and 805R, targeting the V3-V4 hypervariable region of bacteria. The 460-bp amplicon was evaluated by high-throughput sequencing using Illumina MiSeq, followed by bacterial population analysis using QIIME 2.
