Plant immunity involves enemy recognition, signal transduction, and defensive response against pathogens by activating and stimulating the genetic pathways. As pathogens step out of the mechanical barriers, the entire game shift to genetic warriors in plants. The disease resistance mechanism in plants enclaves directly or indirectly involved complex pathways. This study attempted to unlock the genetic elements that contribute to the dieback disease of Dalbergia sissoo, an important timber tree. Using a degenerate oligonucleotide-primed polymerase chain reaction, we have identified differentially expressed gene sequences that showed expression only in resistant sources under dieback challenge. The identified genetic elements (resistance gene analogs or RGAs) were in silico characterized as the contributors of disease-resistance pathways that switch on under dieback challenge. The structural and functional characterization predicted their role in disease resistance pathways. This study paves the way toward resistance genes identification D. sissoo with no genetic record yet.
