Gene expression profiles of normal and injured podocytes induced by immunotoxin.

Podocyte injury is a key event for progressive renal failure. We previously established a mouse model of inducible podocyte injury (NEP25) and demonstrated relentless progression of glomerular injury toward sclerosis. To further investigate molecular events, we performed polysome analysis of intact and injured podocytes utilizing the NEP25 and RiboTag transgenic mice. We show here the expression profiling of normal podocytes and podocytes injured by immunotoxin. RiboTag mice were crossed with Nphs1-Cre mice (designated as Nphs1-RiboTag mice) and podocyte RNA was isolated from podocyte specific polysomes. RNA of Glomerular cells was also obtained from Nphs1-RiboTag mice. Nphs1-RiboTag mice were crossed with NEP25 mice (designated as Nphs1-RiboTag-NEP25). Nphs1-RiboTag-NEP25 mice was injected with LMB2 to induce podocyte injury and podocyte RNA was isolated 4 and 7 days later.
