Diatoms, a prominent category of microalgae, are essential to aquatic ecosystems, functioning as primary producers and indicators of water quality. This study investigates the diversity and ecological importance of diatoms in the deep-water regions of the KHAROON River in RAIPUR CHATTISGARH INDIA, utilizing next-generation sequencing (NGS) technologies. Water samples were systematically collected from key locations along the river to ensure comprehensive representation of the deep-water environments. Genomic DNA was extracted from these samples, and diatom-specific primers were employed to amplify the 18S rRNA and rbcL gene regions. The resulting sequences were analyzed using high-throughput sequencing platforms, allowing for a detailed characterization of diatom diversity. The findings reveal notable differences in diatom community composition, shaped by environmental factors such as nutrient concentrations, water depth, and flow velocity. Bioinformatics analysis identified both cosmopolitan and site-specific diatom taxa, enhancing our understanding of the biogeography and ecological interactions within the Kharoon River. The results emphasize the efficacy of NGS as a robust tool for bio-monitoring and managing water resources in river ecosystems. This research not only broadens our understanding of diatom diversity but also establishes a framework for the application of molecular techniques in the study of freshwater ecosystems. The implications of these findings are significant for biodiversity conservation and the sustainable management of river habitats amid anthropogenic pressures and climate change.
