Sprague Dawley rats were treated with monocrotaline 60 mg/kg by intraperitoneal injection or vehicle control on day 0. On day 23, animals were sacrificed and pulmonary artery endothelial cells (PAECs) were isolated for transcriptomic analysis. To isolate rat PAECs, lung tissue was excised, enzymatically digested, and the resulting single cell suspension was subjected to immunomagnetic bead selection for CD31. CD31+ cells were subjected to a second sorting step using Griffonia simplicifolia isolectin B4 (GS-IB4) and a different CD31 antibody via fluorescence activated cell sorting. GS-IB4+/CD31+ intact cells were analyzed with RNA sequencing to identify differentially expressed genes between monocrotaline PAH and control.
