N6-methyladenosine (m6A), a commonly post-transcriptional modification of eukaryotic mRNAs, playing oncogenic roles or behaving as tumor suppressors in various human cancer types. Medulloblastoma, one of the most common malignant, pediatric brain tumor, which is thought to arise from disruptions in cerebellar development. Amid all 4 subtypes, SHH-subtype medulloblastoma frequently occur in cerebellar hemispheres and more common in children. Here, we uncovered whether the progression of SHH-subtype medulloblastoma also involves RNA m6A regulation. In line with the elevated expression level of METTL3, our direct m6A-seq of SHH-MB samples revealed significant RNA m6A hypermethylation in thousands of genes. Furthermore, we identified that METTL3 could activate SHH signaling pathway in m6A-dependent manner, which further promoted SHH-MB progression.
