5- aminolevulinic acid (5-ALA) is a valuable drug and agricultural precursor, but its microbial production is limited by its close coupling with the biosynthesis of essential heme. Here, we decouple 5-ALA synthesis from heme metabolism in Corynebacterium glutamicum by regulating the activity of porphobilinogen synthase (PBGS, encoded by hemB). Metabonomics analysis confirmed that down-regulation of PBGS mainly inhibited porphyrin biosynthesis and had the least effect on central carbon metabolism.
