Tuft cells and fibroblasts promote thymus regeneration through activation of a type-2 immune response [ex_vivo_ILC2]

The goal of this study was to determine whether thymic innate lymphoid cells type-2 (ILC2) activation during acute thymic involution is mediated by the alarmins IL-25 and IL-33. For this purpose we sorted thymic ILC2 from WT mice, treated them ex vivo with recombinant IL-25/IL-33 or both for 24 hours, and performed bulk RNA Seq. Overall design: ILC2 from thymi of young male WT mice were sorted and grown ex vivo, then treated with rIL-25, rIL-33, both, or vehicle only (PBS) for 24 hours and then mRNA profiles were generated by deep sequencing using the MARSseq method. 3-4 replicates were used.
