The objectives of this study were to characterize bone marrow alteration in the context of kidney disease. Bone marrow (BM) aspirates were collected from patients with chronic kidney disease (CKD). This study included adult patients with CKD (primary FSGS, secondary FSGS, or non-FSGS) who had previously undergone kidney transplant and have developed recurrence in the renal allograft (n=9, age>18 yrs, both genders, all races). Participation in this study was entirely voluntary. We excluded patients who have acute humoral or cell-mediated rejection, thrombocytopenia, leukocytopenia, hematocrit below 20% or a known reaction to Lidocaine. BM aspirates of healthy donors were purchased from Lonza Inc. and used as controls. BM samples were shipped overnight and processed the next day. From these samples, BM mononuclear cells (BM-MNCs) were isolated. The resulting BM-MNCs cells were cryopreserved in freezing medium for this subsequent scRNA-seq analysis. Scipio bioscience Asteria kit (REV-gel technology) and Cytonaut software was used for analysis.
