Rationally Designed Human p16INK4a Mini-Promoter Sequences for Senescence-Targeted Gene Therapy

This dataset contains the nucleotide sequences of five rationally designed human p16INK4a mini-promoter variants. These sequences were designed to overcome the packaging size limitation of adeno-associated virus (AAV) vectors while maintaining or enhancing senescence-specific activity. The dataset includes sequences for p16-0.9kb-mwt, p16-0.9kb-opt1, p16-0.79kb-opt2.1, p16-0.67kb-opt2.2, and p16-0.59kb-opt3.2, in Fasta (.fasta) format.Important Note:These sequences represent the rational design phase of an ongoing undergraduate research project. They are based on comprehensive bioinformatic analyses (including multi-species alignment, CpG island prediction, and transcription factor binding site analysis) and have been designed to test specific hypotheses regarding p16INK4a promoter minimization and optimization. These sequences are computationally designed and have not yet been experimentally validated. Researchers are advised to use them with this understanding.Welcome feedback and collaboration to improve these designs.
