Effect of depletion of HIF-1a, FACT or RNF20/40 on gene expression in hypoxia of MCF7 breast cancer cells

To investigate how HIF-1 recruits FACT and RNF20/40 to mediate histone ubiquitination and facilitate transcription of target genes, we established stable cell lines in which HIF-1, FACT or RNF20/40 has been knocked down by shRNA. We performed gene expression profiling analysis using data obtained from RNA-seq of 4 different cells at 20% and 1% oxygen condition. Overall design: Comparative gene expression profiling analysis of RNA-seq data for MCF7 subclone of NTC, HIF-1a, FACT and RNF20/40 knockdown that were exposed in 20% or 1% oxygen condition for 24 hours.
