Compare staining quality on live PBMCs versus PBMCs pre-fixed with 1.6% PFA for 10min at RT (fixed before surface staining).
Conclusion:
Although the majority of the antigens were not overtly altered in their dynamic ranges, a subset of antigens (including CCR7 and CD11b) showed decreased staining on previously fixed cells. However, manually gated immune cell frequencies from live-stained cells versus cells fixed with PFA before surface staining were nevertheless highly correlated (r = 0.94).
