ApoD modulation mouse cerebellar transcriptome

The lipocalin Apolipoprotein D (ApoD), known to protect the nervous system against oxidative stress (OS) in model organisms, is up-regulated early in the mouse brain in response to the ROS generator paraquat (PQ). However, the processes triggered by this up-regulation have not been explored. We present here a study of the effect of ApoD on the early transcriptional changes upon OS in the mouse cerebellum using microarray profiling. ApoD-KO and transgenic mice over-expressing ApoD in neurons are compared to wild-type controls.

脂钙蛋白载脂蛋白D(lipocalin Apolipoprotein D, ApoD)已被证实可在模式生物中保护神经系统抵御氧化应激(oxidative stress, OS)。当活性氧发生器百草枯(paraquat, PQ)作用于小鼠脑部时,ApoD的表达会在早期被上调,但该上调事件所触发的生物学过程尚未得到探索。本研究采用基因芯片分析(microarray profiling)技术,探究了ApoD对小鼠小脑氧化应激下早期转录变化的影响,实验中将ApoD基因敲除(ApoD-KO)小鼠、神经元过表达ApoD的转基因小鼠与野生型对照小鼠进行了对比分析。
