Cells were sampled at distinct time points (day-1, 3, 6, 12 and 18) and processed for single-cell RNA sequencing (scRNA-seq) to provide a comprehensive profile of dynamic cellular signature during differentiation from human iPSC into spinal cord neural progenitors/precursors. To investigate the therapeutic potential of spNPGs for spinal cord injury (SCI), iPSC-spNPGs were harvested at day12 and used to injected into contusive injury epicenter of non-obese diabetic severe combined immunodeficient (NOD-SCID) mice at 1week post-injury. Then chimeric spinal tissue containing injury lesions and grafted iPSC-spNPG cells were collected for single-nuclei RNA sequencing (snRNA-seq) to confirm the in vivo fate of grafts.
