Mouse macrophages Raw264.7 were infected with M. smegmatis at an MOI = 0.1 for 1 h, washed with PBS to remove extracellular mycobacteria, and the control group continued to culture the infected macrophages for 24 h or 72 h using medium containing PBS.In the experimental group, infected macrophages were treated using different concentrations of cisplatin for 24 or 72h; Macrophages were lysed, and bacteria were collected and quantified by colony forming unit (CFU) assay. CFU experiments showed that cells treated with DDP showed a decreased number of bacteria within macrophages compared with the control group treated with PBS. To dissect the molecular mechanism underlying the reduced number of mycobacteria within macrophages caused by cisplatin, gene expression studies were performed.
