To characterize the functional consequences for CLL development, we generated a targeted Btk gene knockout in Eµ-TCL1-transgenic mice. Strikingly, Btk deletion did not prevent the development of leukemia in TCL1tg/wtBtk-/- mice. To understand the differential signaling events occurring in Btk-deficient versus Btk-proficient CLL cells, we investigated the transcriptomes of Btk-deficient versus wildtype CLL and performed bulk RNA sequencing on total splenocytes isolated from highly leukemic TCL1tg/wtBtkwt/wt (n=4) and TCL1tg/wtBtk-/- (n=4) mice.
