Exploring Resistomes from Viable Isolates in First Nation Community of Manitoba

To enhance our comprehension of the dynamics of antimicrobial resistance within wastewater systems, our study delves into the resistome of these bacterial samples procured from a wastewater treatment plant situated in a First Nation Community, positioned west of Winnipeg, Manitoba, Canada. Utilising a case-control approach in our investigation, we included a site situated upstream from the sampling location which did not encompass the wastewater (treated or untreated) from the corresponding community. Over the period from September 2022 to April 2023, we meticulously collected a total of 58 encircling raw sewage, oxidation lagoon, SAGR, and UV-treated samples. Subsequent to sample acquisition, DNA analysis and whole genome sequencing were carried out, targeting specifically the coliform bacterial components of these aliquots. We harnessed the MinION Sequencer by Oxford Nanopore Technologies for rapid, real-time long-read sequencing. Subsequently, the data was analyzed by using the De-novo assembly tool in the Geneious prime platform.
