Splicing factor mutations are characteristic of myelodysplastic syndromes (MDS) and related myeloid neoplasms and implicated in their pathogenesis, but their roles in the development of MDS have not fully been elucidated. We investigated the consequence of mutant Srsf2 expression under the steady-state condition as well as in a regenerative context using newly generated Vav1-Cre-mediated conditional knock-in mice. We evaluated the effect of mutant Srsf2 on RNA splicing and expression through RNA sequencing of hematopoietic stem and progenitor cells purified from the bone marrow of Srsf2 mutant mice both under the steady-state and after transplantation.
