Halting Cancer Metastasis Through Targeted Adhesion Modulation and Next-Generation Biomaterials

This perspective article, authored by Sami Rashid Mohammed Shibah, an independent Yemeni researcher based in Saudi Arabia (ORCID: 0009-0005-3128-416X, Email: sami0593002781sami@gmail.com), introduces a pioneering anti-metastatic strategy to halt cancer dissemination, the leading cause of cancer mortality. By precisely modulating adhesion molecule dynamics, the approach enhances Cadherin and Integrin-mediated adhesion to anchor cancer cells at their primary site while inhibiting Selectin and IgSF-mediated adhesion to block metastatic colonization. Leveraging state-of-the-art biomaterials—dynamic ECM scaffolds, targeted nanoparticles, ECM stabilizers, and bioinspired peptides—this strategy integrates precision oncology with advanced bioengineering. The manuscript elucidates molecular mechanisms, proposes innovative therapies, and critically evaluates historical challenges (e.g., MMP inhibitor failures) and current barriers, such as tumor heterogeneity and toxicity. A robust roadmap for clinical translation, supported by multi-omics, advanced preclinical models, and smart delivery systems, is outlined.
