The de novo DNA methyltransferase DNMT3C protects male fertility from transposon activity (small RNA-seq)

To allow the characterization of the populations of small RNAs in the testes of mutant animal, small-RNA fraction of total testis RNA was isolated and sequenced. Comparison of small RNA-seq populations from whole testis RNA of mutant Dnmt3C IAP/IAP and control Dnmt3C IAP/WT at 16.5 dpc. A parallel experiment done with Miwi2 KO/WT and KO/KO at the same developmental stages was conducted as a control.为表征突变动物睾丸中的小RNA（small RNA）群体，本研究分离并测序了总睾丸RNA中的小RNA组分。对16.5 dpc（days post coitum，着床后天数）的突变型Dnmt3C IAP/IAP与对照型Dnmt3C IAP/WT个体的全睾丸RNA进行小RNA测序（small RNA-seq），并比较二者的小RNA群体特征。同期在相同发育阶段开展了以Miwi2敲除（knockout, KO）杂合型（KO/WT）与纯合型（KO/KO）个体为对象的平行对照实验。
