Genotyping and Genome Assembly of the Medicinal Plant Anoectochilus roxburghii

Anoectochilus roxburghii is a highly valued medicinal plant in traditional Chinese medicine, recognized for its unique pharmacological properties and broad therapeutic potential. To establish a foundational genomic resource for this species, we performed a de novo assembly of its haplotype-resolved, chromosome-level genome. This was achieved by integrating high-fidelity PacBio long-read sequencing with high-throughput chromosome conformation capture (Hi-C) data. The resulting haplotype assemblies reached sizes of 1.92 Gb and 1.93 Gb, with impressive contig N50 values of 22.72 Mb and 22.17 Mb, respectively. Furthermore, we annotated 26,239 and 26,324 high-confidence protein-coding genes in the two haplotypes. This study provides a chromosome-scale genomic framework that will significantly advance future research into the genetic basis of the plant's medicinal properties and support its conservation and biotechnological application.
