The pestalotiopsis-like taxa, complete with Pseudopestalotiopsis, Pestalotiopsis, Neopestalotiopsis, is a well-known fungal family. The Neopestalotiopsis species have recently been identified as plant pathogens on crops and fruits. There is still a lack of high-quality reference genome of Neopestalotiopsis spp. in germplasm resource evaluation research. In this study, we report a chromosome-level reference genome of N. cubana based on the DNBSEQ platform and PacBio RS II platform. The 3, 627 Mb clean datas and 5, 767 Mb subreads were obtained by DNBSEQ and PacBio RS II platform, separately. The whole genome was assembled through 140 contigs with a total length of 50.40 Mb and a contig N50 length of 0.56 Mb. We obtained a genome size of 50, 399, 148 bp, which consisted of a circular chromosome. In addition, a total of 15, 709 protein-coding genes, and 657 ncRNAs were annotated in the reference genome. The genome sequence data offer a better understanding of the lifestyle strategy and pathogenesis of Neopestalotiopsis species. Our study significantly enriches the species richness of Neopestalotiopsis spp. and provides a basis for follow-up studies.
