The complete genome and methylome of Pseudomonas alcaligenes NEB585, the source of PacI restriction endonuclease, was determined using PacBio sequencing and methylation analysis. We found no methyltransferase component for the PacI endonuclease R-M system. The absence of protective methylation is made possible by the absence of any PacI recognition sites (TTAATTAA) within the genome.
