Chiffon triggers global histone H3 acetylation and expression of developmental genes in Drosophila embryos [ChIP-seq]

Purpose: The goal of this study was to identify H3K14ac levels in chiffon mutant vs WT Method: Drosophila embryos (stage 12-14) was used to ChIP H3K14ac and H3. 1ug of Drosophila chromatin was mixed to 50ng of yeast chromatin (spike-in) and ChIP was performed. Then we generated ChIP libraries using Ovation Ultralow library system (NuGEN) with unique dual indices for multiplexing. Samples were sequenced using Ilumina HiSeq150 next generation sequencer (three biological replicates per genotype: chiffon null mutants flies, and WT: chiffon mutant expressing the transgene chiffon FL.
