We genetically manipulated human bronchial epithelial cells from three non-smoking donors to target genes frequently mutated in lung squamous cell carcinoma. Mutant cells harboured nine combinations of five genetic modifications (including wild-type). Modifications include TP53ko(T), CDKN2Ako(C), KEAP1ko(K), PTENko (P) and SOX2-overexpression (S). Bulk-RNAseq analysis was carried out on total RNA extracted from organotypic air-liquid interphase cultures generated each.
