Single-cell RNA-sequencing of CD45+ immune cells from a kidney transplant biopsy and matched peripheral blood

T cells are central mediators of transplant rejection, yet the differentiation trajectories that sustain this process remain incompletely defined. To delineate the differentiation trajectory of CD8+ T cells, we performed single-cell transcriptomic profiling of matched allograft and peripheral blood mononuclear cell (PBMC) samples obtained from the same patient. Overall design: A renal allograft biopsy was obtained percutaneously under ultrasound guidance using a 14-gauge core biopsy needle. The biopsy tissue was immediately placed on ice and rapidly processed for downstream analysis. Peripheral blood mononuclear cells (PBMCs) were collected from the same subject to enable matched sequencing. CD45+ immune cells from the biopsy and PBMC sample were isolated and profiled by single-cell RNA sequencing.
