Pseudomonas putida, a soil-dwelling bacteria, has become more popular as a chassis for microbial engineering due to its resilient nature and adaptability to a plethora of conditions. To better understand its gene expression changes from the lab to soil, P. putida cultures were grown under lab conditions (Luria broth, LB) and a proxy for the soil environment, soil extracted soluble organic matter (i.e. SESOM), in the exponential and stationary growth phases. RNA from was then extracted and purified, and then libraries were prepared for Illumina sequencing. In addition, further analysis was done to determine the viability of SESOM as a substitute for plain soil.
