Cell migration and invasion are critical factors in tumor progression and metastasis. Wound healing assay and Boyden chamber assay are effective tools for investigating tumor development, as both can measure cell migration rates. Therefore, a simple integrated polydimethylsiloxane (PDMS) device for cell migration assay has been developed, which enables quantitative evaluation of cell migration behavior, particularly for wound healing assays. This integrated device comprises three modules: a cell culture dish, a PDMS chamber, and a wound-generating mold. The PDMS chamber is integrated with the cell culture compartment, allowing six independent experiments to be conducted simultaneously under different stimulatory conditions. To validate the device's functionality, the migration behavior of tumor cells at various time points under different concentrations of fetal bovine serum (FBS) and transforming growth factor-β (TGF-β) was investigated using this device. This device possesses the unique capability of generating wound regions in parallel during cell migration assays, providing a simple and effective platform for studying cell migration assays in biomedical applications.
