RNA-Seq from CA1 pyramidal neurons purified by FACS from 4-week-old mice. CA1 pyramidal neurons were labeled with TdTomato by crossing Camk2a-Cre (B6.Cg-Tg(Camk2a-cre)T29-1Stl/J) and Ai14 (B6.Cg-Gt(ROSA)26Sortm14(CAG-tdTomato)Hze/J mice). The Ai14 reporter line expresses robust tdTomato fluorescence following Cre-mediated recombination. Hippocampi were dissociated from postnatal day 28-32 male Camk2a-Cre +/-, Ai14 +/- mice and TdTomato positive cells isolated by FACS. 3 biological replicates were performed each with cells from 2-4 mice. RNA was extracted from the purified CA1 pyramidal neurons and subjected to RNA-Seq.

本数据集源自通过荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACS)从4周龄小鼠体内纯化得到的CA1锥体神经元的RNA测序(RNA-Seq)数据。通过将Camk2a-Cre(B6.Cg-Tg(Camk2a-cre)T29-1Stl/J)与Ai14(B6.Cg-Gt(ROSA)26Sortm14(CAG-tdTomato)Hze/J)小鼠品系杂交,使CA1锥体神经元被TdTomato荧光蛋白(TdTomato)标记。Ai14报告基因小鼠品系在Cre介导的重组反应后可稳定表达高强度的tdTomato荧光信号。实验从出生后第28至32天的雄性Camk2a-Cre +/-、Ai14 +/-小鼠体内分离海马组织,通过FACS分选获得TdTomato阳性细胞。本实验设置3次生物学重复,每次重复使用2至4只小鼠的细胞。从纯化得到的CA1锥体神经元中提取总RNA,并进行RNA-Seq测序。
