Purpose: RNAseq analyses were conducted to screen for the genes undergoing transcriptional changes either in the liver of high-fat-diet (HFD)-induced obese mice or in the liver of Lepr-deficient db/db mice compared to the livers of the respective control mice Methods: C57BL/6 wild-type male mice were fed on high-fat diet (HFD) or a low-fat diet (NCD) for 18 weeks starting from 6 weeks of age, and the livers were collected at 24 weeks of age at ad libitum-fed condition.Misty/misty or db/db were sacrificed at ad libitum-fed condition at 10weeks and the liver was collected. Results: 2079 genes and 1085 genes were identified in high-fat-diet fed mice and db/db mice, respectively. Liver mRNA profiles of obese mouse moldels.

研究目的:本研究开展RNA测序(RNA-seq)分析,旨在筛选相较于对应对照组小鼠肝脏,高脂饮食(high-fat diet, HFD)诱导的肥胖小鼠肝脏以及瘦素受体缺陷型db/db小鼠肝脏中发生转录水平变化的基因。 实验方法:选取6周龄的C57BL/6野生型雄性小鼠,分别饲喂HFD与普通低脂饲料饮食(normal chow diet, NCD),自6周龄起持续饲喂18周后,于24周龄自由进食状态下采集肝脏组织;将Misty/Misty小鼠与db/db小鼠于10周龄自由进食状态下处死,采集肝脏组织。 实验结果:在HFD组小鼠与db/db小鼠中,分别鉴定出2079个与1085个差异表达基因。本数据集涵盖肥胖小鼠模型的肝脏mRNA表达谱。
