Dispersive solid phase extraction of antisense oligonucleotides with the use of amino acid based adsorbents

Data have been deposited on the development of a method of dispersive solid-phase extraction of antisense oligonucleotides using adsorbents modified with amino acid molecules. The impact of the composition of solvents used for adsorption and desorption was tested (pH, salt type, salt concentration, organic solvent type, and content in the solution). All tested adsorbents allowed for the efficient recovery of oligonucleotides. Nevertheless, adsorbent with bonded methionine showed the highest efficiency, probably due to the presence of a sulfur atom. The recovery depended on the structure of the antisense oligonucleotide. It was higher for shorter oligonucleotides and modified ones (phosphorothioate, 2′-O-methyl, 2′-O-(2-methoxyethyl) and both phosphorothioate and 2′-O-methyl in one oligonucleotide). The results revealed that the recovery also increases with increasing hydrophobicity of the modification. Data on the characteristics of the synthesized adsorbents, kinetics of oligonucleotide adsorption, sorption capacity were also deposited. Moreover, the results of applying the developed extraction method to human plasma samples are also included.
