Differential miRNA expression of healthy and inflammatory periodontal ligament stem cells-derived exosomes

To further explore the specific expressed miRNA in healthy and inflammatory periodontal ligament stem cells (PDLSCs)-derived exosomes, we extracted their exosomal RNA for miRNA microarray assay. The culture supernatants of healthy PDLSCs (H-PDLSCs, obtained from healthy periodontal ligament tissue) and inflammatory PDLSCs (P-PDLSCs, obtained from periodontitis periodontal ligament tissue; L-PDLSCs, H-PDLSCs treated with lipopolysaccharide for 7 days) were used for exosomes extraction (H-Exo, P-Exo and L-Exo) by using ultracentrifuge. The supernatants used for exosomes extraction mixed equal amounts of 5 individuals H-PDLSCs or P-PDLSCs culture supernatants and the source of cells used for L-Exo extraction was the same as H-Exo; n=5. P-EXO vs H-EXO; L-EXO vs H-EXO; P-EXO vs L-EXO.
