SMG1, a Nonsense-Mediated mRNA Decay (NMD) regulator, as a therapeutic target in multiple myeloma

RNA expression was profiled in B-cell lymphoma cell lines treated with CC0483115 (CC-115) using strand-specific RNA-sequencing. The following cell lines were tested: GRANTA (mantle cell lymphoma, resistant to CC-115), JEKO (mantle cell lymphoma, sensitive), Jiyoye (Burkitt lymphoma, resistant), RAMOS (Burkitt lymphoma, sensitive), RPMI8226 (multiple myeloma (MM), sensitive), and U-266 (MM, resistant). Overall design: Strand-specific RNA-seq was performed on 6 B-cell lymphoma cell lines, untreated and treated with CC-115 Various B-cell lymphoma cell lines were cultured, treated with CC-115 or vehicle (DMSO), and harvested. Samples were transferred to NeoGenomics for library preparation withTruSeq Stranded mRNA kits and paired-end sequencing on Illumina HiSeq 2500.
