Genome-wide titration of gene expression in M. tuberculosis and M. smegmatis

Genome-wide CRISPRi libraries in M. tuberculosis H37Rv, M. tuberculosis HN878, and M. smegmatis mc2-155 were grown in the presence and absence of ATc to induce CRISPR interference. Titration of expression was achieved through the use of single-guide RNAs of different efficiency. Libraries were sequenced at approximately 0, 2.5, 5, 7.5, 10, 15, 20, 25 and 30 generations of cumulative growth.
