The textbook view holds that transcriptionally inactive heterochromatin forms condensed domains, whereas active euchromatin is open and accessible. Using advanced nanoscopic imaging, we revealed condensed euchromatic domains labeled by H3.3-HaloTag in living human cells, challenging the long-standing notion that euchromatin is largely open. These condensed euchromatic domains may contribute to higher-order gene regulation. In this project, we purified H3.3-HaloTag-labeled euchromatin from HCT116 cells, sequenced it, and mapped the corresponding genomic regions.
