Sumoylation alters the transcriptional activity of many of the transcription factors known to regulate CSCs/TICs and epithelial-mesenchymal transition (EMT). To identify transcription factor involved in the growth effects mediated by SUMO inhibition, Chromatin immunoprecipitation DNA-sequencing (ChIP-seq) was used to evaluate genomic occupancy by SUMO1 and Etv1. Overall design: To investigate SUMOylation inhibition, we established Ube2iF/F cell line to achieve complete conditional knockout by Adneovirus-Cre. We then performed Adenovirus comparing to control to knockout SUMOylation before ChIP-seq. We can then measure the quantitative occupancy of SUMO1 and Etv1 throughout the genome.
